While many fans may expect Pep Guardiola‘s side to run riot, the supercomputer – from OLBG – predicts Spurs to knock them out.
The two sides last met at the beginning of December, with Tottenham holding the treble-winners to a 3-3 draw.
City thought they had won the match following Jack Grealish’s 81st minute goal, but Dejan Kulusevski scored a late equaliser to clinch a point in the Premier League.
Neither team will be hoping for a draw this time around, with both managers desperate to avoid a replay.
In another potential shock, League Two side Wrexham are expected to see off Blackburn Rovers.
Elsewhere, Manchester United and Liverpool are predicted to comfortably beat opponents Newport County and Norwich respectively.
Chelsea‘s clash with Aston Villa is expected to be fiercely contested, with the Blues set to scrape through.
Meanwhile, Ipswich are predicted to beat the lowest-ranked side left in the competition, Maidstone United.

The Tractor Boys are currently flying high in the Championship table, with Maidstone fourth in the National League South.
Championship leaders Leicester City are also expected to progress with a win over Birmingham.
And Premier League sides Bournemouth, Wolves and Nottingham Forest are among those expected to win this weekend.
But in some of the all-Premier League fixtures we could be set for more shocks.
The supercomputer predicts Sheffield United to beat Brighton at Bramall Lane and Fulham to beat Newcastle at Craven Cottage.
Sheffield United are currently rock bottom of the Premier League table, so a good cup run will certainly help to lift the spirits of fans.
In the other all-Championship fixtures, Watford are set to beat Southampton and Leeds are predicted to defeat Plymouth Argyle at Elland Road.
